Driven by the need for more sensitivity and the opportunity for wide applicability of nucleic acid testing in point of need environments, there is a strong market desire to have a fast, robust, nucleic acid detection technology married to the speed and convenience of a lateral flow technology.

The detection of nucleic acid product in point of need environments has been made difficult by a variety of factors including the technical difficulties associated with the most commonly used amplification technology, PCR. The main challenges are in sample preparation and control of assay specificity and reproducibility. Recombinase Polymerase Amplification (RPA), developed by ASM Scientific, Cambridge,UK, represents a significant step forward in the reduction of the complexity of nucleic acid amplification methods to the point where they can be applied in point of need environments. The appropriate application of lateral flow technology, with some modifications, can assist in bringing this key application directly to the point of need.

LFIA’s have not yet been widely applied in this application. Traditional LFIA’s have often suffered from limitations in sensitivity and reproducibility. These limitations are mainly due to the use of traditional manfacturing practices, materials, labels, and visual detection systems. In recent years, advances in these areas have allowed for significantly improved performance in lateral flow assays. This makes it possible to apply LFIA’s in areas, such as nucleic acid detection, that may demand more sensitivity and reproducibility than traditional lateral flow assays are typically considered capable of.

DCN’s range of assay readers, coupled with our ability to develop and manufacture very high sensitivity fluorescent, magnetic or visual assays provide a way to quickly produce a range of assays that can address specific needs within this market. Our ability to develop and design devices, plastics, sample handling and preparation systems, and integrate everything into a complete assay system that can be rapidly applied at the point of need make DCN the ideal partner for your assays in the this area of application.

For a detailed discussion of market opportunities and appropriate technologies for lateral flow and flow through market segments, see DCN’s Technology and Market Report on the point of need diagnostics market, entitled Pushing Lateral Flow to the Limits. Contact us for information on how to order this report.

Introduction

Agricultural applications of lateral flow and flow through technology are numerous, ranging, from testing of grains, grapes and other crops for the presence of naturally occurring toxins, to assessment of quality of grains, determination of fungal or bacterial contamination of crops, to assessment of crop quality and identification of genetically modified (GMO) crops.

Lateral flow and flow through assays are ideal for this type of application, given their ability to handle a wide variety of sample types and perform many sample handling tasks on the device. Filtration, concentration and removal of cross-reactive elements are all important in these types of tests. Lateral flow and flow through assays can be specifically designed to handle all of these elements. High sensitivity and ease of use by untrained users are also critical factors in uptake of assays in this field. These are traits of lateral flow and flow through assays, particularly when integrated with appropriate handheld or benchtop reader systems based on fluorescence, magnetics or visual labels (colloidal gold or colored latex).

Numerous opportunities are available in this market, taking advantage of the sensitivity, specificity, ease of use and the growing ability for quantification with these assay formats. Among the largest are indentification of Genetically Modified Crops, through either phenotypic or genotypic assays. DCN has worked with companies developing isothermal DNA and RNA amplification methods, which are ideal for linking to lateral flow or flow through rapid assays. Get more information on DCN’s work in the area of Link to Nucleic Acid Lateral Flow.

Introduction

This section relates to food safety diagnostics involving bacterial contamination of raw materials, manufacturing equipment and manufactured product. The primary industries involved in this sector are catering and food service establishments, food and beverage manufacturers, meat processing and packing plants, dairies and breweries.

Foodborne transmission of pathogenic and toxigenic microorganisms has been a recognized hazard for decades. Food safety is evaluated in terms of acceptable levels of risk and is enhanced by systematically minimizing contamination at every point from food production and processing to distribution, preparation, and consumption. The food test menu includes a wide variety of organisms. The most common contaminants include: Salmonella, Shigella, Listeria monocytogenes, Escherichia coli O157:H7, Campylobacter jejuni, Vibrio parahaemolyticus, Yersinia enterocolitica, and others.

In the past 3-5 years food safety issues and concerns for public health have lead to more stringent legislation in food safety requirements. Legislation has produced increased demand for pathogen and toxin tests in just about every segment of the food production industry – processed food, meats, poultry, beverages, and dairy; and by all major food producers worldwide.

The market for food diagnostics is highly diversified and still developing. The food and beverage sector (food) is the largest segment within the industrial rapid test market. Overall, more than 600 million tests were performed, worldwide in 2005, with a market value of $1 billion. This sector is expected to see annual growth of 5% for the next five years.

In 2005/06 Lateral flow tests for food and beverage testing accounted for only 9% of all immunoassays used in this sector ($290 million, worldwide, in 2005). Improvements in performance and integration of new paradigms, such as nucleic acid lateral flow detection coupled with isothermal amplification methods will result in increased penetration of the market by rapid tests.

Introduction

There is growing interest in rapid, point of need tests for potential biowarfare agents, which may include infectious agents such as anthrax, or toxins such as Staph Enterotoxin B or ricin. The potential variety of delivery systems and vectors (food, water supplies, air, surfaces in public areas) and the need for rapid, accurate evaluation of threats and of potentially infected individuals in times of extreme stress and possible mass exposure make this an area with very unique needs. Lateral flow assays, coupled with reader systems represent one of the best platforms for this type of testing due to their speed, ease of use, flexibility in terms of matrices that the assays can handle, ease of interpretation, speed of development and ease of large scale manufacture in time of need.

This market is driven largely by government demands and as a result accurate figures are difficult to source. There is no doubt that it is a growing segment, and one where this technology can be readily and appropriately applied.

DCN’s range of assay readers, coupled with our ability to develop and manufacture very high sensitivity fluorescent, magnetic or visual assays provide a way to quickly produce a range of assays that can address specific needs within this market. Our ability to develop and design devices, plastics, sample handling and preparation systems, and integrate everything into a complete assay system make DCN the ideal partner for your assays in the Biowarfare / Biodefense area.

The primary consumer based lateral flow assay is the over the counter pregnancy test, which constitutes a large component of the $2.1 billion dollar lateral flow test market. Other consumer based applications are growing in popularity, including tests for contaminants in food and beverages (e.g. caffeine), and at-home or in-store alcohol testing. Home drugs of abuse testing, and home-use infectious disease testing are growing applications in many markets. The lateral flow and flow through assay formats lend themselves well to consumer applications for a variety of reasons, including ease of use for untrained users, accurate results, and, if linked to an at-home or in-store reader system, can produce clear results that are interpreted accurately by the reader virtually 100% of the time. The linkage of reader systems to these tests also allow for new paradigms for use. Data can be stored on the readers, and used to monitor the patient’s status, either in real time, via connectivity, or at a later date after downloading by a medical professional. Depending on the application, if the reader is connected to the internet via USB or Bluetooth, the data can be stored, evaluated and used for certain commercial purposes, such as the delivery of notifications or coupons for the purchase of certain other products (e.g. flu remedies linked to influenza testing). Accurate market numbers for current consumer tests are not readily available, as the pregnancy test element of this market currently dwarfs other applications, however it is a new and growing area of interest for many companies.

Approximately 60% of immunoassays for small and large animals are performed in reference laboratories. In 2005, LF-type tests represent some 39% of the veterinary rapid immunoassay and ELISA tests ($450 million) performed worldwide. These LF tests generated revenues of $175 million, worldwide and with 9% CAGR will reach $270 million in 2010.

The market is highly fragmented with different tests used for specific animal species. Some 60% of the LF vet market is occupied by tests for household pets. These tests include: tests for anemia, cancer, endocrine function; viral pathogens: canine adenovirus type-1 (CAV-1), calicivirus (CaCV), adenovirus, coronavirus, parvovirus, rabies, rotavirus, West Nile Virus, etc.; and bacteria and parasites: E. coli, cryptosporidia, heartworm, hookworm, leishmania, leptospirosis, Lyme disease, roundworm and tapeworm.

Large animal veterinary medicine is becoming an important facet of public health, worldwide. Public health and food supply, everywhere, are at constant risk from accidental or malicious introduction of animal diseases that destroy animal stock or can be transferred to humans. The threats include obvious ones like foot and mouth disease (FMD), as well as agents that affect animals and humans, such as bovine tuberculosis and paratuberculosis, avian influenza, anthrax, bovine spongiform encephalopathy (BSE) and West Nile virus.

DCN’s broad experience in the development of assays in this area of application, coupled with our ability to design devices specifically for these applications and integrate readers as appropriate, make DCN your ideal partner for the development or manufacture of assays specifically for veterinary use.
